Heat shock proteins (Hsp) were opened in the second part of XX century as single group of proteins [Ritossa, 1962;Tissneres, 1974]. Hsp were found in cells of all organisms, including archeobacterium. Intracellular expression of Hsp70 is necessary for cells survival and conservation theirs function under influence of exogenous chemical and physical agents on organism [Margulis, Guzhova, 2000].  The task of the present investigation is to study  sleep and wakefulness states characteristics under consecutive Hsp70 (without endotoxin)  and  cholinergic agonist carbachol microinjections  in Nucleus reticularis pontis oralis (NRPO). It is now well known that NRPO is critical structure, including in regulation of Rapid Eye Movement Sleep (REMS) [Xi et al, 2002].

Five adult pigeons (Columba livia) were housed at a free movement condition with ambient temperature 23-24 оС, on a 12:12 h light/dark cycle, with ad libitum access to food and water. Computer recording (with epoch 1 s during 24 h) of electroencephalogram (EEG), electrooculogram (EOG), electromyogram (EMG), electrocardiogram (ECG), brain temperature (Тbr) and foot skin temperature (Тfs) was used for an analysis of sleep-wakefulness states. Bilateral microinjections of cholinergic agonist carbachol (0.2 mkg/0.1 mkl) were performed in one hour after microinjections of Hsp70 (without endotoxin ;0.4 mkg / 0.3 mkl) into mesencephalic part of NRPO through cannulas implanted by coordinate atlas [Karten, Hodos, 1967] at the onset of dark phase. The control animals were injected with saline.    

Findings is that after microinjections of Hsp70 the microinjections of carbachol did not induced dramatic an increase in REMS total time (TT) during first two hours of dark phase. A decrease in REMS TT and wakefulness TT were obtained in following hours. But non REM sleep (non REMS) TT is increase. Recently we found the following effects: the microinjections of carbachol was induced a significant rise in the number of REMS episodes and REMS TT (by 12-15 times) in comparison with control during first two hours of dark phase [Guselnukova et al., 2002]; the microinjections of Hsp70 were induced a significant rise in non REMS TT and a decrease in wakefulness TT in 7-12 hours of dark phase [Guselnikova , 2005]. Hsp70 is likely to influence on synaptic transmission of acetylcholine and prevent from its drastic liberation induced by carbachol microinjections.

Data obtained show that Hsp 70 possess neuroprotective properties.
